Spira doesn't have any reasons to attempt pulling the sword in the first place, in fact, he feels that he's the least worthy of the group to handle such an important artifact, but after seeing Shiva fail to pull out the sword, he'd ask her one thing in particular, if she felt anything in particular, (mainly referring to the burning sensation) after getting an affirmative response, Spira would share that, he too, felt the burning sensation and would tug on the handle of the sword, to see if his theory holds. If it doesn't move, he postulates his theory; "I think that the sword burns, or tries to burn, people that aren't worthy to wield it. Therefore, I think we should all (referring just to the PC characters here) touch the sword and narrow it down from there."

Concerning the moss: Spira picks a piece of the moss, roughly a pinch of it, himself, and after eyeing it closely, sticks his tongue to it, he doesn't eat it, just tastes it. He then tries to make something like nori sheets out of the moss.
